My favourite comedy series (other than Python) is Futurama. Always laugh with it. Always.
My favorite show (aside from Flying Cirus) would have to be either Mr Show or, somewhat predictabley, Arrested Development. It's hard to recommend those to people since most have seen them, but when you find a MS or AD virgin, introducing them to the sheer comedic genius of either of those shows is so VERY satisfying.
Hard to vote against 'Seinfeld' either way. A long and consistent run, and I'll still watch the reruns without hesitation.

Okay, it's really hard to top the Monty Python bunch (omg - I SO LOVED their 'Flying Circus' program)...
BUT, you just can't beat British humor, so I will say definitely MR. BEAN!!! (Rowan Atkins rocks)

Really off-the-wall, funny stuff, and pretty groundbreaking for sketch humor. The way the sketches segue into the next, and sometimes come full circle to where the episode began, is pretty cool.

It still holds up 13 years after that overdose, bringing a smile regularly.
Runners up would be the original Bob Newhart, and Barney Miller for US shows. Probably the too-brief Fawlty Towers, or Alan Partridge shows in a UK context (unless we can count QI!)
It had some moments that were less successful than I suspect they'd like, but so much of it still stands up, many years late. I've always been impressed by how much of it (and how much of the best of it) was done as in-character improvisation. The fact that it wasn't a pure comedy, but had serious drama as its other side, highlighted the lighter moments terrifically. I doubt we shall ever see its like again, and hope to be proved wrong.
Best current comedy? My Name is Earl. Trailer Trash meets 12 Steps. An inspired and surprisingly gentle satire.
All those characters in the twelve episodes were just like real life people most of us have known, displaying their hilarious oddities .

I found it hilarious... at the time
Flying Circus and Kids in the Hall were very formative though.
Plus... all episodes are available to watch for free online all over the world, yay for creators who understand how the internet works!
Monty Python's comedic sensibilities updated in the 90s.
Through all its ups and downs, there's always been plenty to make me laugh. So many legends of comedy have passed through its ranks.
